GRF1 Antibody Overview

GRF1 antibodies target Ras-GRF1, a 155 kDa guanine nucleotide exchange factor (GEF) that activates Ras and Rac GTPases by catalyzing GDP-to-GTP exchange . Ras-GRF1 contains distinct domains:

  • CDC25 homology domain (Ras-GEF activity)

  • IQ motif (calmodulin binding)

  • Dbl homology (DH) and pleckstrin homology (PH) domains (Rac-GEF activity)

These antibodies are essential for detecting GRF1 expression in tissues like pancreatic islets, brain, and testis .

Pancreatic β-Cell Development

GRF1 antibodies revealed reduced β-cell mass in GRF1 knockout mice due to impaired proliferation and neogenesis :

  • 30% reduction in serum insulin levels

  • Defective IGF-I signaling: Akt and Erk activation failure in β-cells

  • Normal insulin secretion but reduced β-cell area (Figure 6B)
