The AIP1-1 antibody (e.g., clone F-3, catalog #sc-365921) is a mouse monoclonal IgG2aκ antibody developed to detect AIP1 across species (mouse, rat, human) . It recognizes epitopes in the N-terminal region of AIP1, enabling applications in western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and ELISA .
AIP1 (DAB2IP) is a Ras-GAP protein with dual roles:
Anti-Angiogenic: Inhibits VEGFR2-PI3K signaling, suppressing pathological angiogenesis .
Pro-Apoptotic: Activates ASK1 by displacing 14-3-3 inhibitors, promoting TNF-α–induced apoptosis .
Tumor Suppression: Downregulated in prostate cancer and leukemia; inhibits Ras, PI3K/Akt, and NF-κB pathways .
Isoforms:
AIP1A: Full-length, anti-inflammatory, localized to cytoplasmic membranes .
AIP1B: Truncated (lacks PH domain), pro-inflammatory, mitochondrial-localized .
Retinal Neovascularization: AIP1-KO mice showed enhanced VEGF-induced angiogenesis, reversed by AIP1 overexpression .
Tumor Microenvironment: AIP1 in stromal cells suppresses VEGFR2 signaling, reducing EMT and metastasis .
ASK1 Activation: AIP1 mediates TNF-α–induced ASK1 activation by dissociating 14-3-3 inhibitors .
Isoform Switch: Chronic inflammation shifts AIP1A (anti-inflammatory) to AIP1B (pro-inflammatory), exacerbating vascular disease .
Prostate Cancer: Low AIP1 correlates with tumor progression; AIP1 reexpression inhibits Ras and PI3K/Akt pathways .
Leukemia: Chromosomal aberrations in AIP1 are linked to acute myeloid leukemia .
