Function
ANGPT1 (Angiopoietin-1) is a protein that binds and activates the TEK/TIE2 receptor. This activation occurs by inducing dimerization and tyrosine phosphorylation of the receptor. ANGPT1 plays a critical role in regulating various aspects of vascular function, including angiogenesis, endothelial cell survival, proliferation, migration, adhesion, and cell spreading. It also influences the organization of the actin cytoskeleton and the maintenance of vascular quiescence. During embryogenesis, ANGPT1 is essential for normal angiogenesis and heart development. Postnatally, its role in angiogenesis is context-dependent, acting as either an activator or inhibitor. ANGPT1 promotes vascular stability and inhibits angiogenesis in quiescent vessels, where endothelial cells exhibit tight junctions. In these vessels, ANGPT1 oligomers recruit TEK to cell-cell contacts, forming complexes with TEK molecules from adjacent cells. This leads to the preferential activation of phosphatidylinositol 3-kinase and the AKT1 signaling pathways. Conversely, in migrating endothelial cells lacking cell-cell adhesions, ANGPT1 recruits TEK to contacts with the extracellular matrix, forming focal adhesion complexes. This process activates PTK2/FAK and downstream kinases MAPK1/ERK2 and MAPK3/ERK1, ultimately stimulating sprouting angiogenesis. ANGPT1 also mediates blood vessel maturation and stability, suggesting a role in endothelial developmental processes distinct from that of VEGF. It appears to play a crucial role in mediating reciprocal interactions between the endothelium and its surrounding matrix and mesenchyme.
