CADM2 antibodies are widely used in:

ApplicationTechniqueKey Findings
Cancer BiologyWestern Blot, IHCCADM2 downregulation correlates with glioma , colorectal , and lung cancer progression .
NeuroscienceImmunofluorescenceRegulates synaptic adhesion and neural circuit formation .
Cell MigrationTranswell/Wound HealingInhibits metastasis via EMT suppression in HCC and NSCLC .

Oncogenic Suppression

  • Glioma: CADM2 overexpression inhibits proliferation (57.6% reduction in U87 cells) and metastasis by downregulating β-catenin and upregulating E-cadherin .

  • Colorectal Cancer: Low CADM2 correlates with advanced tumor stage and poor survival (HR = 2.1, p < 0.001) .

  • Lung Cancer: Silencing CADM2 enhances EMT, increasing brain metastasis risk (OR = 3.2) .

Mechanistic Insights

  • Cell Cycle Regulation: Reduces cyclin D1 and CDK4/2 expression, arresting G1/S transition .

  • EMT Modulation: Suppresses vimentin and Slug while enhancing E-cadherin in HCC and NSCLC .

Clinical Implications

  • Biomarker Potential: CADM2 expression predicts survival in glioma (median OS: 22 vs. 14 months, high vs. low) and CRC (AUC = 0.84) .

  • Therapeutic Target: Restoring CADM2 inhibits FAK/AKT signaling, reducing HCC metastasis .

Limitations and Future Directions

  • Antibody Specificity: Variability in isoform detection remains a challenge .

  • Translational Gaps: Preclinical models require validation in human trials for therapeutic applications .

Target Background

Function
CADM2, also known as Cell Adhesion Molecule 2, is an adhesion molecule that participates in both homo- and heterophilic interactions with other members of the nectin-like family. These interactions lead to cell aggregation, playing a crucial role in synapse organization by providing regulated trans-synaptic adhesion. Notably, CADM2 exhibits a preferential binding affinity for oligodendrocytes.
