Description

CFHR2 Protein Overview

CFHR2 is a 50 kDa plasma glycoprotein composed of four short consensus repeat (SCR) domains. Key features include:

PropertyDetail
Gene locationChromosome 1q32 (within the CFHR gene cluster)
StructureHomodimer formed via SCR1-2 domains; C-terminal SCR3-4 binds C3b/C3d
Plasma concentration~50 µg/mL
Key functionsInhibits C3 convertase and terminal complement complex (TCC) formation

CFHR2 shares 93% sequence identity with SCR19-20 of factor H but lacks competition for C3b binding, enabling cooperative regulation with factor H .

Research Applications of CFHR2 Antibodies

Key experimental uses include:

  • Immunohistochemistry: Localizing CFHR2 in glomerular dense deposits (C3 glomerulopathy)

  • Functional assays: Quantifying CFHR2-mediated inhibition of C3 convertase activity

  • Disease studies: Investigating CFHR2 polymorphisms (rs3790414) in AMD cohorts

  • Protein-protein interaction: Analyzing CFHR2 binding to C3b/C3d via surface plasmon resonance

Key Research Findings Using CFHR2 Antibodies

Recent studies utilizing these antibodies have revealed:

  • Mechanistic insights: CFHR2 binds C3b without displacing factor H, allowing simultaneous regulation of complement activation .

  • Disease associations:

    • CFHR2 deficiency correlates with increased AMD risk (OR: 1.7–2.3)

    • CFHR2 accumulates in renal biopsies of C3 glomerulopathy patients

  • Structural analysis: SCR3-4 domains mediate C3d binding with KD = 12 nM, while SCR1-2 enable dimerization .

Target Background

Function
CFHR2 plays a role in complement regulation. Its dimerized forms exhibit a strong affinity for tissue-bound complement fragments, effectively competing with the physiological complement inhibitor CFH. CFHR2 can associate with lipoproteins and may contribute to lipid metabolism.
Gene References Into Functions
  1. Research suggests an association between the CFHR2-rs2986127 variant and acute anterior uveitis (AAU) diagnosis. PMID: 27306586
  2. Next-generation sequencing of the CFH region identified potentially functional variants (missense, splice site, and indel) on the four common haplotypes. Notably, we found no expression of any of the five CFH-related genes in the retina or RPE/Choroid/Sclera, unlike the liver, which serves as the primary source of circulating proteins, including CFHR2. PMID: 27196323
  3. CFHR2 likely works in conjunction with factor H, as CFHR2 inhibits convertases while simultaneously facilitating factor H-mediated convertase degradation by factor I. PMID: 24260121
  4. A hybrid CFHR2-CFHR5 plasma protein, resulting from a chromosomal deletion mutation, stabilizes the C3 convertase and reduces factor H-mediated convertase decay. PMID: 24334459
