Function
CREB3L2 is a transcription factor that plays a crucial role in the unfolded protein response (UPR). In the absence of endoplasmic reticulum (ER) stress, CREB3L2 is embedded in ER membranes, with its N-terminal DNA-binding and transcription activation domains facing the cytosol. Upon ER stress, CREB3L2 is transported to the Golgi apparatus, where it undergoes site-specific cleavage by resident proteases S1P/mbtps1 and S2P/mbtps2. The resulting N-terminal cytosolic domain translocates to the nucleus, initiating transcription of specific target genes. CREB3L2 is essential for chondrogenesis and may protect neuroblastoma cells from ER stress-induced apoptosis. In vitro, CREB3L2 directly binds to the cAMP response element (CRE) site, activating transcription of target genes.
