Description

Research Applications

CTNNA2 antibodies are utilized across diverse experimental workflows:

Key Applications

  • Western Blot (WB): Detects CTNNA2 in lysates from human, mouse, and rat tissues (e.g., MCF7 cells, neuroblastoma models) .

  • Immunohistochemistry (IHC): Visualizes protein localization in brain, pancreas, and tumor tissues .

  • Functional Studies:

    • Silencing or overexpressing CTNNA2 modulates laminin-dependent adhesion and invasiveness in head and neck squamous cell carcinoma (HNSCC) .

    • Low CTNNA2 expression correlates with neuroblastoma relapse in non-MYCN-amplified patients .

Cancer Biology

  • Tumor Suppression: CTNNA2 loss promotes β-catenin phosphorylation, enhancing Wnt pathway activity and tumor invasion . In neuroblastoma, reduced CTNNA2 levels predict poor relapse-free survival .

  • Mutation Impact: Mutants like CTNNA2-S132Y increase Matrigel invasion in HNSCC, mimicking pathogenic phenotypes .

Neurological Disorders

CTNNA2 regulates synaptic plasticity and neuronal migration, with disruptions linked to cortical dysplasia .

Performance Notes

  • Storage: Stable at -20°C in glycerol-containing buffers .

  • Validation: Supported by knockdown/overexpression studies and structural homology .

Recent Advances (2023–2025)

  • Structural Insights: AlphaFold predictions reveal CTNNA2’s actin-binding domain architecture, aiding antibody epitope mapping .

  • CPTAC Assays: Targeted proteomics (CPTAC-1756) quantify CTNNA2 in ovarian cancer, showing reproducible detection with <10% CV .

  • Therapeutic Exploration: Antibodies like ab76015 enable studies on CTNNA2’s role in suppressing Arp2/3-mediated actin polymerization, relevant to metastasis .

Limitations and Future Directions

  • Species Restriction: Most antibodies lack cross-reactivity beyond human/mouse/rat .

  • Functional Variants: Impact of splice isoforms (e.g., NP_001269526.1) on antibody specificity requires further validation .

Target Background

Function
CTNNA2 antibody functions as a linker between cadherin adhesion receptors and the cytoskeleton, playing a crucial role in regulating cell-cell adhesion and differentiation within the nervous system. It is essential for proper regulation of cortical neuronal migration and neurite growth. CTNNA2 acts as a negative regulator of Arp2/3 complex activity and Arp2/3-mediated actin polymerization, thus suppressing excessive actin branching that could hinder neurite growth and stability. This antibody regulates morphological plasticity of synapses and cerebellar and hippocampal lamination during development, and contributes to the control of startle modulation.
