CUL4A and CUL4B share 83% sequence identity but exhibit distinct subcellular localization and functional roles :

  • CUL4A: Localizes to both the nucleus and cytoplasm, forming the CRL4A complex with DDB1 and ROC1. It recruits substrates via WD40-repeat proteins (DCAFs) for ubiquitination .

  • CUL4B: Primarily nuclear, facilitating DDB1 nuclear import independently of DDB2 .

Both proteins are neddylated for activation and interact with COP9 signalosome (CSN) and CAND1 to regulate substrate specificity .

Function
CUL4 is a core component of the CUL4-RBX1-CDD (COP10-DDB1a-DET1) E3 ubiquitin-protein ligase complex. This complex plays a crucial role in mediating the ubiquitination and subsequent proteasomal degradation of target proteins. CUL4's involvement in the CDD complex contributes to light-mediated developmental control. Notably, it may repress photomorphogenesis by enhancing COP1 E3 ubiquitin-protein ligase activity. CUL4 collaborates with the CUL4-DDB1-COP1-SPA E3 ubiquitin-protein ligase complexes in regulating photomorphogenesis and flowering time. Furthermore, CUL4 is a constituent of the CUL4-RBX1-DDB1-PRL1 E3 ubiquitin-protein ligase complex, which mediates the ubiquitination and degradation of AKIN10. Additionally, it is a component of the CUL4-RBX1-DDB1-DWA1/DWA2 E3 ubiquitin-protein ligase complex, acting as a negative regulator in abscisic acid (ABA) signaling, potentially targeting ABI5 for degradation.
