Target Background

Function
DDX3X, a multifunctional ATP-dependent RNA helicase, plays a crucial role in RNA remodeling. While not essential for RNA unwinding, it binds to RNA in a concentration-dependent manner, facilitating annealing between complementary RNA strands. This process is further regulated by ATP, which reduces RNA binding and consequently diminishes annealing. DDX3X is involved in a wide range of cellular processes, not necessarily requiring its ATPase/helicase catalytic activities. These processes include the regulation of transcription and translation initiation, participation in innate immunity, and involvement in both stress and inflammatory responses. Notably, DDX3X promotes liquid-liquid phase separation of P granules, a process crucial for intracellular organization and stress granule assembly. It is essential for embryonic development and plays a role in sexual cell fate determination by negatively regulating the translation of the sex determining protein tra-2. DDX3X may have a protective role in responding to heat and oxidative stress and may negatively regulate the extrinsic apoptotic signaling pathway via death domain receptors. Additionally, it may be involved in mitotic chromosome segregation.
