DOF3 Antibody is a transcriptional activator that binds specifically to the DNA consensus core sequence 5'-AAAG-3', also known as the prolamin box. It can activate the expression of genes encoding for the seed storage proteins glutelin, prolamin, and globulin. DOF3 functions synergistically with RISBZ/BZIP58 to positively regulate the expression of numerous seed storage proteins. Additionally, it collaborates with RISBZ1/BZIP58 to positively regulate the expression of certain metabolic enzymes, such as alanine aminotransferase and pyruvate phosphate dikinase, which are expressed during seed development. DOF3 also works in conjunction with RISBZ1/BZIP58 to positively regulate genes crucial for the development of aleurone layers and the glutelin GLUD-1 gene in the endosperm of developing seeds. Furthermore, DOF3 can activate the expression of the bifunctional lysine-degrading enzyme, lysine ketoglutarate reductase/saccharopine dehydrogenase (LKR/SDH), a key regulator of free lysine content in plants. During seed germination, DOF3 participates in the gibberellin-mediated activation of the alpha-amylase AMY1.1/AMY1A gene.
