Pol β is a repair polymerase that plays a crucial role in base-excision repair. It possesses 5'-deoxyribose-5-phosphate lyase (dRP lyase) activity, enabling it to remove the 5' sugar phosphate. Additionally, Pol β functions as a DNA polymerase, adding a single nucleotide to the 3' end of the resulting single-nucleotide gap. Its DNA synthesis during 'gap-filling' occurs in a stepwise distributive manner, distinct from the processive nature observed in other DNA polymerases.
