Description

Research Applications

The At3g61080 Antibody is utilized in diverse experimental contexts:

  • Gene Expression Analysis: Quantification of AT3G61080 mRNA/protein levels via immunoblotting or immunofluorescence .

  • Epigenetic Studies: Investigation of gene silencing and paramutation in polyploid Arabidopsis lines .

  • Cellular Localization: Tracking protein distribution in tissues like gynoecia, septa, or ovules .

Key Findings:

  • In ntt stk double mutants, AT3G61080 expression is reduced, indicating regulation by NO TRANSMITTING TRACT (NTT) and STK proteins .

  • The antibody aids in studying cell wall metabolism and transmitting tract development in plant reproductive tissues .

Gene Regulation

  • NTT-STK Interaction: AT3G61080 is part of a core coexpression network regulated by NTT and STK. ChIP-qPCR experiments confirmed promoter binding of STK to AT3G61080, suggesting direct transcriptional control .

  • Polyploidy Studies: In autotetraploid Arabidopsis, AT3G61080 expression patterns were analyzed alongside epigenetic changes during paramutation .

Methodological Utility

  • Immunoblotting: Used to validate protein knockdown in mutants .

  • Immunolocalization: Combined with LM21 antibody (mannan detection) to study cell wall polysaccharide dynamics .

Target Background

Function
This antibody initiates a process leading to the deglycation of proteins. It specifically phosphorylates low-molecular-mass and protein-bound erythrulosamines and ribulosamines, but not fructosamines or psicosamines, on the third carbon of the sugar moiety. The resulting protein-bound erythrulosamine 3-phosphates and ribulosamine 3-phosphates are unstable and decompose under physiological conditions.
