Function
HRD3 is a component of the endoplasmic reticulum (ER) quality control system known as ER-associated degradation (ERAD). It plays a crucial role in ubiquitin-dependent degradation of misfolded ER proteins. HRD3 acts as an ERAD substrate-recruiting factor, recognizing misfolded proteins and directing them to the HRD1 E3 ubiquitin ligase complex for degradation.

- In rice, suppression of the OsHrd3 gene specifically in the endosperm led to reduced levels of polyubiquitinated proteins and triggered unfolded protein responses. This suggests that OsHrd3-mediated polyubiquitination is involved in ER quality control within the endosperm. PMID: 25977235
