Introduction
IL19, a member of the IL10 cytokine subfamily, is primarily expressed in monocytes. Upon binding to the IL20 receptor complex, IL19 activates STAT3 signaling. In mice, a similar cytokine exhibits pro-inflammatory properties by upregulating IL6 and TNF-alpha expression and inducing apoptosis. This suggests a potential role for IL19 in inflammatory responses. Notably, alternative splicing of the IL19 transcript gives rise to distinct isoforms.
Description
Recombinant human IL19, expressed in HEK293 cells, is a single, glycosylated polypeptide chain encompassing amino acids 25-177. With a molecular weight of 19.1 kDa, it comprises 164 amino acids. A 6-amino acid His-tag is fused to the C-terminus to facilitate purification, which is achieved using proprietary chromatographic techniques.

Formulation
The IL19 protein solution (0.5 mg/ml) is formulated in a buffer containing 10% glycerol and Phosphate-Buffered Saline (pH 7.4).
Stability
For short-term storage (2-4 weeks), the product can be stored at 4°C. For extended storage, freezing at -20°C is recommended. The addition of a carrier protein (0.1% HSA or BSA) is advisable for long-term storage. Repeated freeze-thaw cycles should be avoided.
Purity
The purity of the IL19 protein is greater than 95.0%, as determined by SDS-PAGE analysis.

Expression and Production

IL-19 is preferentially expressed in monocytes, macrophages, and T and B lymphocytes . It is produced by B cells and monocytes and binds to the interleukin 20 receptor complex to activate STAT3 signaling . The production of IL-19 is up-regulated in resting monocytes following stimulation with granulocyte-macrophage colony-stimulating factor (GM-CSF) or lipopolysaccharide (LPS) .

Function and Signaling

IL-19 plays a significant role in the immune response by promoting T helper 2 (Th2) cell-mediated immune responses .
