Description

Experimental Use Cases

The INTS8 antibody has been validated in multiple studies:

ApplicationKey FindingsReference
Western Blot (WB)Detects INTS8 in MCF7 and HeLa cell lysates; confirms overexpression in cholangiocarcinoma .
Immunohistochemistry (IHC)Highlights elevated INTS8 protein levels in human intrahepatic cholangiocarcinoma (CHOL) tissues compared to normal samples .
Immunofluorescence (IF)Localizes INTS8 in nuclear compartments of HeLa cells, consistent with its transcriptional role .

Recommended Dilutions

ApplicationDilution Range
Western Blot (WB)1:200–1:1000
Immunoprecipitation (IP)0.5–4.0 µg per 1–3 mg lysate
Immunofluorescence (IF)1:10–1:100

Technical Notes

  • Optimization: Titration is critical due to batch-specific variability .

  • Controls: Use HIBE (normal biliary epithelial) and CHOL cell lines (HCCC-9810, RBE) for comparative studies .

Functional Roles

  • Transcriptional Regulation: INTS8 is a core component of the Integrator complex, essential for snRNA 3'-end processing and transcriptional termination .

  • Cancer Progression: Drives epithelial-to-mesenchymal transition (EMT) via TGF-β signaling in HCC and CHOL .

  • Neurodevelopmental Disorders: Mutations in INTS8 destabilize the Integrator complex, impairing neuronal differentiation and mRNA processing .

Clinical Implications

Cancer TypeAssociation with INTS8
Cholangiocarcinoma (CHOL)Overexpression linked to advanced stage and immune evasion .
Pan-CancerHigh INTS8 levels predict poor disease-specific survival .

Target Background

Function
INTS8 is a component of the Integrator complex. This complex is involved in the transcription of small nuclear RNAs (snRNAs) U1 and U2, and their 3'-box-dependent processing. The Integrator complex associates with the C-terminal domain (CTD) of the RNA polymerase II largest subunit (POLR2A) and is recruited to the U1 and U2 snRNA genes.
Gene References Into Functions
  1. Mutations in INTS8 have been linked to a severe neurodevelopmental syndrome. These mutations lead to instability of the Integrator complex and impaired function in vitro. PMID: 28542170
