Target Background

Function
This antibody targets Homoserine Kinase (HSK), an enzyme that catalyzes the ATP-dependent phosphorylation of L-homoserine to L-homoserine phosphate. It exhibits substrate specificity for L-homoserine, excluding other substrates such as D-serine, L-serine, D-threonine, L-threonine, galactose, and D-homoserine *in vitro*. HSK is implicated in the plant's susceptibility to the downy mildew pathogen *Hyaloperonospora parasitica*.
Gene References Into Functions
  • Studies of the *dmr1* (downy mildew resistance 1) mutant allele in Arabidopsis have revealed varying effects on disease resistance. The *dmr1-2* allele showed increased resistance to *Fusarium culmorum* and *F. graminearum* silique infection, along with decreased rosette leaf colonization. In contrast, the *dmr1-1* allele exhibited reduced rosette leaf colonization but maintained wild-type silique resistance. PMID: 25432266
  • Research suggests that pathogen resistance mechanisms involving impaired PMR4 (callose synthase 12) and DMR1 (homoserine kinase) are conserved across Arabidopsis and tomato. PMID: 23818978
  • Overexpression of HSK did not significantly affect the levels of soluble homoserine phosphate, methionine, or threonine/methionine. PMID: 15703056
  • Analysis of six independent *dmr1* mutants, each with a distinct amino acid substitution in the HSK protein, indicates that homoserine accumulation within the chloroplast triggers a novel form of downy mildew resistance. PMID: 19622802
