Target Background

Function
LAT2, also known as NTAL or LAB, is a transmembrane adaptor protein involved in the signaling pathways of various immune cells. In mast cells, LAT2 plays a role in FCER1 (high affinity immunoglobulin epsilon receptor)-mediated signaling. Additionally, it may participate in BCR (B-cell antigen receptor)-mediated signaling in B-cells and FCGR1 (high affinity immunoglobulin gamma Fc receptor I)-mediated signaling in myeloid cells. LAT2 acts as a crucial link, connecting the activation of these receptors and their associated kinases to downstream intracellular events through the recruitment of GRB2.
Gene References Into Functions
  1. Overexpression of LAT2 has been associated with poor prognosis in cancer, correlating with tumor grade, proliferative potential, and angiogenesis. It also plays a role in mediating the intracellular transport of anticancer agents. PMID: 28815339
  2. The AML1/ETO target gene LAT2 interferes with the differentiation of normal hematopoietic precursor cells. PMID: 24456692
  3. LAT2 serves as an early mediator of the anti-leukemic activity of alkylphospholipids and arsenic trioxide. Consequently, LAT2 holds potential as a therapeutic target for cancer treatment. PMID: 23001822
  4. NTAL functions as a tumor suppressor, enhancing the proximal signaling of leukemic blasts. ERK, a key downstream molecule, is responsible for the biological effects of TCR signaling. PMID: 22269118
  5. LAB possesses the ability to modulate signaling pathways in virtually all types of leukocytes. PMID: 20643813
  6. LAT2 protein (NTAL) participates in the activation of the c-Met-Grb2-ERK-cPLA2 signaling cascade during the early stages of H. pylori infection. PMID: 19879355
  7. Research suggests that NTAL (non-T cell activation linker) might be a structural and possibly functional homologue of LAT (linker for activation of T cells) in non-T cells. PMID: 12486104
  8. LAB links BCR engagement to downstream signaling pathways. PMID: 12514734
  9. LAB is primarily phosphorylated on three membrane-distal tyrosines, Tyr(136), Tyr(193), and Tyr(233). Mutations in these tyrosines disrupt Grb2 binding and LAB function. PMID: 14722116
  10. LAB resembles a LAT molecule that is unable to bind phospholipase C-gamma1. PMID: 15153499
  11. The NTAL/LAB is expressed in various immune cells. PMID: 15745852
  12. NTAL acts as a negative regulator of TNF-alpha and IL-8 production following stimulation via TREM-1. PMID: 17277102
  13. Findings indicate that NTAL may be differentially utilized by specific receptors for relaying alternative signals, suggesting a greater flexibility in the function of TRAPs than previously understood. PMID: 17993265
  14. Data demonstrate that natural killer cell immunoreceptor tyrosine-based activation motifs preferentially utilize a signaling cassette regulated by the interplay between LAT and LAB. PMID: 18645037
  15. Expression of LAT2 in T lineage leukemias separates them into two subgroups with distinct prognoses in response to prednisone therapy in children with acute lymphoblastic leukemia. PMID: 19183565

Involvement In Disease
LAT2 is located in the Williams-Beuren syndrome (WBS) critical region. WBS results from a hemizygous deletion of several genes on chromosome 7q11.23, thought to arise as a consequence of unequal crossing over between highly homologous low-copy repeat sequences flanking the deleted region. Haploinsufficiency of LAT2 may be the cause of certain cardiovascular and musculo-skeletal abnormalities observed in the disease.
Subcellular Location
Cell membrane; Single-pass type III membrane protein. Note=Present in lipid rafts.
Tissue Specificity
Highly expressed in spleen, peripheral blood lymphocytes, and germinal centers of lymph nodes. Also expressed in placenta, lung, pancreas and small intestine. Present in B-cells, NK cells and monocytes. Absent from T-cells (at protein level).
