Description

2.1. Role in Cancer Biology

  • Overexpression in Tumors: MCM5 is upregulated in lung adenocarcinoma (LUAD), oral squamous cell carcinoma (OSCC), and glioblastoma (GBM), correlating with poor prognosis .

    • In LUAD, MCM5 interacts with histone deacetylase 1 (HDAC1), promoting metastasis and reducing patient survival .

    • OSCC studies show MCM5 knockdown reduces cell proliferation by inducing G2/M phase arrest and downregulating cyclin E/CDK2 .

  • Therapeutic Target: High MCM5 expression in GBM enhances chemoresistance to temozolomide (TMZ), suggesting its utility as a therapeutic target .

Table 1: MCM5 Expression in Cancer Types

Cancer TypeKey FindingSource
Lung AdenocarcinomaCo-expression with HDAC1 predicts poor survival; promotes invasion/metastasis
GlioblastomaDrives TMZ resistance; linked to CREB1, NRF1 transcription factors
Oral Squamous CellSilencing reduces proliferation via p21 upregulation and cyclin E downregulation

2.2. Viral Replication Mechanisms

  • HIV-1 Interaction: MCM5 incorporates into HIV-1 virions and modulates reverse transcription. Both depletion and overexpression of MCM5 impair viral replication, indicating a biphasic role .

2.3. Transcriptional Regulation

  • Stat1-Mediated Signaling: MCM5 binds to IFN-γ-activated Stat1 target promoters (e.g., IRF-1) to facilitate transcription, independent of its DNA replication role .

Clinical and Diagnostic Relevance

  • Biomarker Potential:

    • MCM5 urine assays are validated for detecting high-risk non-muscle-invasive bladder cancer recurrence .

    • In laryngeal squamous cell carcinoma (LSCC), MCM5 expression correlates with Ki-67, a standard proliferation marker .

Target Background

Function
MCM-5 antibody targets the MCM-5 protein, a component of the MCM2-7 complex (MCM complex). This complex serves as the replicative helicase, crucial for the initiation and elongation of DNA replication during the eukaryotic cell cycle. The MCM2-7 complex functions on a 'once per cell cycle' basis. The active ATPase sites within the MCM2-7 ring are formed by the interaction surfaces of adjacent subunits. This interaction leads to the formation of a conserved arginine finger motif in trans, relative to the ATP-binding site of the Walker A box of the adjacent subunit. While all six ATPase active sites contribute to the complex helicase activity, their contributions likely differ.
