Product Specs

Introduction
MS4A1, found on the surface of all B-cells, is an activated-glycosylated phosphoprotein. Its expression starts during the pro-B stage and increases until maturity to ensure a robust B-cell immune response, particularly against T-independent antigens. MS4A1 is involved in B cell microenvironmental interactions, the regulation of B-cell activation and proliferation, and acts as a calcium channel within the cell membrane.
Description
Produced in Sf9 Baculovirus cells, Recombinant Human MS4A1 is a single, glycosylated polypeptide chain. It consists of 97 amino acids (210-297a.a.), has a molecular mass of 11.1 kDa, and appears between 18-28 kDa on SDS-PAGE due to glycosylation. The protein includes a 9 amino acid His tag at the C-terminus and is purified using proprietary chromatographic techniques.
Physical Appearance
A clear, sterile-filtered solution.
Formulation
The MS4A1 protein solution has a concentration of 0.5mg/ml and contains phosphate-buffered saline at a pH of 7.4 with 10% glycerol.
Stability
For short-term storage (2-4 weeks), keep at 4°C. For longer periods, freeze at -20°C. Adding a carrier protein (0.1% HSA or BSA) is suggested for long-term storage. Avoid repeated freeze-thaw cycles.
Purity
Purity is greater than 90% as determined by SDS-PAGE analysis.

Product Science Overview

Structure and Function

MS4A1 is a B-lymphocyte-specific membrane protein that plays a crucial role in the regulation of cellular calcium influx necessary for the development, differentiation, and activation of B-lymphocytes . The protein contains four highly conserved transmembrane domains, flanked by N- and C-terminal cytoplasmic regions . It acts as a store-operated calcium (SOC) channel component, promoting calcium influx after activation by the B-cell receptor (BCR) .

Expression and Localization

The MS4A1 gene is localized to chromosome 11q12, among a cluster of family members . It is widely expressed during B-cell ontogeny, from early pre-B-cell developmental stages until final differentiation into plasma cells . The unique expression patterns of MS4A1 among hematopoietic cells and nonlymphoid tissues highlight its importance in the immune system .

Clinical Significance

MS4A1 is implicated in various diseases, including Common Variable Immunodeficiency (CVID) and Diffuse Large B-Cell Lymphoma (DLBCL) . The protein is a target for therapeutic antibodies, such as Rituximab, used in the treatment of B-cell malignancies . Its role in B-cell activation and calcium influx regulation makes it a critical component in immune response and a valuable target for immunotherapy .

Research and Applications

Recombinant MS4A1 is used in research to study B-cell biology, immune response, and the development of therapeutic antibodies. It is also utilized in various assays and diagnostic applications to understand its function and role in diseases .
