MSRB1 is a methionine-sulfoxide reductase that specifically catalyzes the reduction of methionine (R)-sulfoxide back to methionine. While methionine oxidation often arises from random oxidation during oxidative stress, it can also serve as a post-translational modification on specific residues. MSRB1 acts as a regulator of actin assembly by reducing methionine (R)-sulfoxide residues on actin, which are modified by MICALs (MICAL1, MICAL2, or MICAL3). This reduction promotes filament repolymerization. MSRB1 plays a role in innate immunity by reducing oxidized actin, leading to actin repolymerization in macrophages.
