Applications of NCBP3 Antibody

NCBP3 antibodies are widely used in:

  • Western Blotting: Detecting endogenous NCBP3 expression in cell lysates .

Study FocusKey FindingsCitation
Antiviral DefenseNCBP3-deficient cells show impaired cytokine production and increased viral replication. Mice lacking NCBP3 exhibit severe lung pathology after influenza A infection.
Hypoxia ResponseNCBP3 scaffolds METTL3 and eIF4A2 to enhance m6A RNA methylation and translation under hypoxic stress in cardiomyocytes.
mRNA ExportNCBP3 associates with the exon junction complex (EJC) and TREX machinery, promoting nuclear export of multi-exonic mRNAs.
Translation RegulationNCBP3 competes with ZC3H18 for RNA binding, stabilizing transcripts and supporting polysome assembly.

Biological Significance of NCBP3

NCBP3 antibodies have illuminated its non-redundant roles in:

  • Viral Infections: NCBP3 ensures cytokine mRNA translation, critical for antiviral immunity .

  • Hypoxic Stress: Facilitates METTL3-mediated m6A methylation to enhance cardiac gene translation .

  • Cancer and Disease: Linked to mRNA export defects in transformed cells and viral susceptibility .

Challenges and Future Directions

  • Functional Redundancy: NCBP3 partially overlaps with NCBP2 in mRNA export, complicating phenotype interpretation .

  • Context-Dependent Roles: NCBP3’s impact on translation varies by cell type and stress condition .

  • Therapeutic Potential: Targeting NCBP3 pathways could modulate antiviral responses or hypoxia adaptation in disease .

Target Background

Function
NCBP3 antibody associates with NCBP1/CBP80 to form an alternative cap-binding complex (CBC). This complex plays a crucial role in mRNA export. NCBP3 acts as an adapter protein, linking capped RNAs (m7GpppG-capped RNA) to NCBP1/CBP80. Unlike the conventional CBC, which contains NCBP2 and binds both small nuclear RNA (snRNA) and messenger RNA (mRNA), the alternative CBC with NCBP3 does not bind snRNA. Instead, it specifically associates with mRNA, participating solely in mRNA export.
