OAS1 Antibody: Role in Cancer and Immunology

OAS1 is an interferon-stimulated gene (ISG) involved in antiviral responses and tumor immunity. Recent studies highlight its prognostic and immunological significance in pan-cancer analyses .

Key Research Findings:

  • Prognostic Value:

    • High OAS1 expression correlates with poor survival outcomes in liver hepatocellular carcinoma (LIHC), pancreatic adenocarcinoma (PAAD), and head and neck squamous cell carcinoma (HNSC) .

    • Conversely, elevated OAS1 levels are associated with favorable prognoses in breast invasive carcinoma (BRCA) and kidney renal clear cell carcinoma (KIRC) .

  • Immune Microenvironment:

    • OAS1 expression positively correlates with immune infiltration scores (stromal, immune, and ESTIMATE scores) in most cancers, indicating its role in tumor-immune interactions .

    • Strong associations with M2 macrophage infiltration (pro-tumorigenic) were observed in PAAD, LIHC, HNSC, and BRCA (e.g., CD163 co-expression: R = 0.66 in PAAD) .

  • Therapeutic Potential:

    • OAS1 outperformed standard models in predicting immunotherapy efficacy in melanoma (AUC = 0.75) .

Data Table: OAS1 Expression and Prognosis in Selected Cancers

Cancer TypePrognostic AssociationImmune Correlation (M2 Macrophages)
Pancreatic (PAAD)Poor survivalCD163: R = 0.66
Liver (LIHC)Poor survivalCD163: R = 0.54
Breast (BRCA)Favorable survivalCD163: R = 0.61

OBF-1 Antibody: Role in B Cell Differentiation

OBF-1 (Oct-Binding Factor 1) is a transcriptional coactivator critical for antibody-secreting cell (ASC) differentiation .

Key Research Findings:

  • Genetic Regulation:

    • OBF-1 deficiency blocks ASC differentiation by failing to induce Blimp-1/prdm1 and repress bcl-6, Pax5, and AID .

    • Loss of OBF-1 reduces Syndecan-1+ plasma cells by >90% in T cell-dependent cultures but not in T cell-independent conditions .

  • Functional Impact:

    • IgM secretion remains near-normal in OBF-1−/− ASCs, but IgG1 production is impaired, suggesting isotype-specific regulatory roles .

Data Table: OBF-1 Dependency in B Cell Differentiation

ConditionSyndecan-1+ Cells (%)Antibody Secretion (vs. Wild-Type)

T cell-independent80–90%IgM/IgG1: Normal

Anti-NGF Antibodies: Therapeutic Applications

While unrelated to OAF1, monoclonal antibodies targeting nerve growth factor (NGF) demonstrate clinical relevance in pain management .

Key Research Findings:

  • DS002 (Anti-NGF mAb):

    • Phase I trials showed favorable safety (t½ = 176–294 hours) and no immunogenicity in healthy subjects .

    • Preclinical efficacy in osteoarthritis (OA) pain and cancer pain models .

  • Efficacy in OA:

    • Anti-NGF antibodies (e.g., tanezumab) significantly reduce WOMAC pain scores (SMD = -0.31) and improve physical function (SMD = -0.36) .

Target Background

Function
The PIP2-OAF1 heterodimer functions as a transcriptional activator, inducing the transcription of genes involved in fatty acid beta-oxidation. This process, known as oleic acid induction, occurs when cells utilize fatty acids as their sole carbon source. The heterodimer specifically recognizes and binds to the oleate response element (ORE), also referred to as the peroxisome box, located in the promoter region of numerous genes. These genes include CTA1, FOX1 to FOX3, FAA2, PAS8, PAS10, and others, encoding peroxisomal proteins. The ORE consists of two inverted CGG triplets separated by 14 to 18 intervening nucleotides. OAF1 acts as the sensor for oleate, inhibiting PIP2 activity in non-inducing conditions. Its activity is repressed by glucose.
