PABN1 plays a crucial role in the formation of mRNA precursors (pre-mRNA) by catalyzing the addition of a poly(A) tail, typically composed of 200-250 nucleotides, to the upstream cleavage product. It acts as a stimulator of poly(A) polymerase (PAPOLA), enhancing the processivity of the poly(A) tail elongation reaction and regulating the length of the tail. PABN1 increases the affinity of poly(A) polymerase for RNA. Furthermore, it exhibits a high affinity for both poly(A) and poly(G) sequences. This binding activity may contribute to the protection of the poly(A) tail from degradation.
