Function
PECAM1 (Platelet Endothelial Cell Adhesion Molecule-1, also known as CD31) is a cell adhesion molecule crucial for leukocyte transendothelial migration (TEM) under most inflammatory conditions. Tyrosine 690 (Tyr-690) plays a critical role in TEM, facilitating efficient PECAM1 trafficking to and from the lateral border recycling compartment (LBRC). Tyr-690 is also essential for LBRC membrane targeting around migrating leukocytes. Trans-homophilic interactions contribute to endothelial cell-cell adhesion via cell junctions. Heterophilic interactions with CD177 facilitate neutrophil transendothelial migration. Homophilic PECAM1 ligation prevents macrophage-mediated phagocytosis of viable neighboring leukocytes by transmitting a detachment signal. Conversely, it promotes macrophage-mediated phagocytosis of apoptotic leukocytes by tethering them to phagocytic cells; this detachment signal appears disabled in apoptotic leukocytes. PECAM1 modulates bradykinin receptor BDKRB2 activation and regulates bradykinin- and hyperosmotic shock-induced ERK1/2 activation in endothelial cells. It is implicated in atherosclerosis susceptibility but does not offer protection against apoptosis.
