Target Background

Function

PHB proteins are essential for embryonic development, playing a critical role in somatic and germline differentiation within the larval gonad. A deficiency in PHB proteins leads to impaired mitochondrial biogenesis in body wall muscle cells. These proteins are also required for the clearance of paternal mitochondria following fertilization.

  1. Research has identified prohibitin 2 (PHB2), an inner mitochondrial membrane protein, as a key mitophagy receptor. PHB2 facilitates the targeting of mitochondria for autophagic degradation by binding to the autophagosomal membrane-associated protein LC3 via an LC3-interaction region (LIR) domain. This binding occurs following mitochondrial depolarization and proteasome-dependent outer membrane rupture. PMID: 28017329
