Function
PHB proteins are essential for embryonic development, playing a critical role in somatic and germline differentiation within the larval gonad. A deficiency in PHB proteins leads to impaired mitochondrial biogenesis in body wall muscle cells. These proteins are also required for the clearance of paternal mitochondria following fertilization.

- Research has identified prohibitin 2 (PHB2), an inner mitochondrial membrane protein, as a key mitophagy receptor. PHB2 facilitates the targeting of mitochondria for autophagic degradation by binding to the autophagosomal membrane-associated protein LC3 via an LC3-interaction region (LIR) domain. This binding occurs following mitochondrial depolarization and proteasome-dependent outer membrane rupture. PMID: 28017329
