Target Background

Function
PHO4 is a transcriptional activator that plays a critical role in regulating the expression of repressible phosphatases under phosphate starvation conditions. It binds to the upstream activating sequence (UAS) of several phosphatase-encoding PHO genes. Under high-phosphate conditions, PHO4 is inhibited by the cyclin-CDK complex PHO80-PHO85.

  2. Further analysis of the inducible PHO5 gene demonstrates that topoisomerases are essential for Pho4p transcription factor binding to the PHO5 promoter. This binding is crucial for promoter nucleosome removal during gene activation. PMID: 23284296
  3. Research suggests that RPL12 has an extra-ribosomal function in modulating the transcription of genes that require Pho4p activation. PMID: 21469193
  4. A pathway controlled by Pho80-Pho85 kinase and Pho4 transcription factor can generate three distinct transcriptional outputs. PMID: 14624238
  5. These findings establish a G1 checkpoint adaptation pathway where Pho85 mediates Pho4 downregulation and Sic1 degradation, ultimately releasing Cdc28 activity and promoting the onset of the S phase. PMID: 16964260
  6. Pho4 binding to the SNZ1/SNO1 promoter is associated with changes in chromatin structure. Rpd3 histone deacetylase is required for proper timing of SNZ1/SNO1 expression, while Asf1 histone chaperone is essential for their expression. PMID: 18408055
  7. Pho4 activates certain stress response genes, is required for G1 arrest induced by DNA damage, and remains present in the nucleus even under high-phosphate conditions to regulate gene transcription. PMID: 19108609
