Target Background

Function
Estrogen receptor beta (ESR2) is a nuclear hormone receptor that binds estrogens with an affinity similar to that of ESR1/ER-alpha. It activates the expression of reporter genes containing estrogen response elements (ERE) in an estrogen-dependent manner. However, ESR2 lacks ligand binding ability and exhibits minimal or negligible ERE binding activity, leading to a loss of ligand-dependent transactivation capability.
Gene References Into Functions
  • A study in a Mexican population revealed an association between the rs1256032 polymorphism of the estrogen receptor beta gene and a reduced risk of developing type 2 diabetes. PMID: 29666032
  • Researchers identified a homozygous ESR2 variant, c.541_543del p.(Asn181del) in the highly conserved DNA-binding domain of ER-beta, in an individual with syndromic 46,XY DSD. Additionally, two heterozygous missense variants, c.251G>T p.(Gly84Val) and c.1277T>G p.(Leu426Arg), located in the N-terminus and the ligand-binding domain of ER-beta, were found in unrelated, nonsyndromic 46,XY DSD cases. PMID: 29261182
