Function
The POB3 antibody targets a component of the FACT (Facilitates Chromatin Transcription) complex, a critical chromatin remodeling factor involved in various DNA-dependent processes, including mRNA elongation, DNA replication, and DNA repair. The FACT complex acts as a histone chaperone, dynamically modulating nucleosome structure. During transcription elongation, FACT facilitates the passage of RNA polymerase II by promoting the dissociation of one histone H2A-H2B dimer from the nucleosome, subsequently enabling the reestablishment of the nucleosome after polymerase transit. This process promotes transcription elongation by suppressing cryptic transcription initiation. Furthermore, FACT plays a role in establishing transcription initiation complexes and enhances the binding of SPT15/TBP to the TATA box. In collaboration with replication factor-A protein (RPA), FACT may contribute to nucleosome deposition during DNA replication.
