Pot-1 Antibody targets a telomeric DNA-binding protein that exhibits high specificity for single-stranded C-rich repeat sequences, particularly the 5'-GCCTAA-3' sequence. This binding can occur at either the 5' or 3' telomeric end. Pot-1 plays a crucial role in safeguarding the 5' end of the C-rich strand within the telomere. It acts redundantly with Pot-2 to negatively regulate telomere extension mediated by telomerase. Additionally, Pot-1 regulates telomere length through the alternative lengthening of telomeres (ALT) pathway, an independent mechanism from telomerase. Through its interaction with Sun-1, Pot-1 anchors telomeres to the nuclear envelope in embryonic development.
