Description

Charybdotoxin (ChTX) is a 37 amino acid neurotoxin derived from the venom of the scorpion Leiurus quinquestriatus hebraeus . It is known to block calcium-activated potassium channels, causing hyperexcitability of the nervous system.

Synthesis Analysis

Charybdotoxin has been synthesized using an Fmoc-tBu protocol . The synthetic native molecule was found to be equipotent with the natural toxin in inhibiting the human erythrocyte Ca2±dependent K+ channel.

The molecular structure of ChTX has been determined using two-dimensional nuclear magnetic resonance (2-D NMR) spectroscopy . The protein possesses a small triple-stranded antiparallel beta sheet linked to a short helix by two disulfides and to an extended fragment by one disulfide.
