Role in Cancer and Metastasis

PXN promotes tumor migration and invasion by serving as a scaffold for integrin-mediated signaling. Phosphorylation at Y31/Y118 by PTK6 activates RAC1 via CRK, enhancing metastatic potential . Increased phospho-Y88 PXN correlates with colon cancer progression .

Mechanotransduction and Phosphorylation

  • ULK1/2 kinases phosphorylate PXN at S32/S119 in response to mechanical stress, inhibiting actin polymerization and focal adhesion (FA) assembly .
