Description

Functional Characteristics

Recombinant hbb2 demonstrates oxygen-binding properties modulated by organic phosphates and pH:

  • GTP Sensitivity: GTP reduces oxygen affinity by 70% and eliminates the reverse Bohr effect (where oxygen affinity increases with acidity) .

  • Root Effect: Acidification induces oxygen release (PO₂ increases by 7–8× in rete mirabile), critical for swimbladder gas secretion .

  • CO₂ Transport: Aquaporins in gas gland cells facilitate CO₂ diffusion, enhancing proton release and Root effect activation .

Key functional metrics:

  • P₅₀ (O₂ partial pressure at 50% saturation): 3.5 mmHg (pH 7.4, no GTP) → 12.1 mmHg (with GTP) .

  • Hill coefficient: 1.2–1.5, indicating low cooperativity .

Genomic and Evolutionary Context

hbb2 is part of a teleost-specific hemoglobin diversification driven by whole-genome duplication (WGD):

  • Gene Clusters: Teleost hemoglobin genes reside in two chromosomal regions:

    • MN cluster (flanked by mpg and nprl3) .

    • LA cluster (flanked by lcmt1 and aqp8) .

  • Evolutionary Adaptations:

    • Inversion events in tetraodontid fish (e.g., fugu) repositioned hbb2 homologs, altering gene expression patterns .

    • Retention of paralogous hbb genes post-WGD enabled subfunctionalization for hypoxia tolerance .

Research Applications

Recombinant hbb2 is used to study:

  1. Allosteric Regulation: Mechanisms of GTP and pH effects on oxygen binding .

  2. Comparative Physiology: Evolutionary adaptations in deep-diving vs. shallow-water fish .

  3. Biotechnological Models: Design of synthetic oxygen carriers inspired by reverse Bohr effect mechanisms .

Product Specs

Form
Lyophilized powder. We will ship the format we have in stock. If you have special format requirements, please note them when ordering, and we will fulfill your request.
Lead Time
Delivery times vary by purchase method and location. Consult your local distributor for specific delivery times. All proteins are shipped with standard blue ice packs. For dry ice shipment, contact us in advance; additional fees apply.
Notes
Avoid repeated freezing and thawing. Store working aliquots at 4°C for up to one week.
Reconstitution
Briefly centrifuge the vial before opening to collect contents at the bottom. Reconstitute the protein in sterile deionized water to a concentration of 0.1-1.0 mg/mL. We recommend adding 5-50% glycerol (final concentration) and aliquoting for long-term storage at -20°C/-80°C. Our default final glycerol concentration is 50%.
Shelf Life
Shelf life depends on several factors, including storage conditions, buffer components, storage temperature, and protein stability. Generally, the liquid form has a shelf life of 6 months at -20°C/-80°C, while the lyophilized form has a shelf life of 12 months at -20°C/-80°C.
Storage Condition
Store at -20°C/-80°C upon receiving. Aliquot for multiple uses. Avoid repeated freeze-thaw cycles.
Tag Info
The tag type will be determined during the manufacturing process. If you require a specific tag type, please inform us, and we will prioritize developing it.
