Description

Production and Purification

Recombinant NuoK is produced in E. coli expression systems, followed by affinity chromatography . Critical parameters include:

  • Expression Vector: Optimized for high-yield soluble protein .

  • Storage Buffer: Tris-based buffer with 50% glycerol, pH adjusted for stability .

  • Purity: >85% confirmed via SDS-PAGE .

Reconstitution Guidelines:

  • Lyophilized protein is reconstituted in sterile deionized water (0.1–1.0 mg/mL) .

  • Glycerol (5–50%) is added for long-term storage at -20°C/-80°C .

Biochemical Applications

  • Respiratory Studies: Used to dissect electron transport mechanisms in β-proteobacteria .

  • Drug Target Screening: NDH-1 is a potential target for antimicrobial agents due to its role in pathogen bioenergetics .

  • Enzyme Engineering: Serves as a template for optimizing quinone reductase activity in synthetic biology .

Stability and Handling

  • Storage: -20°C/-80°C in 50% glycerol; avoid freeze-thaw cycles .

  • Activity Loss: <10% after 6 months at -80°C .

Research Gaps

  • Structural Data: No high-resolution structures for Azoarcus NuoK limit mechanistic insights.

  • Kinetic Parameters: Michaelis constants (Kₘ) for NADH/quinones remain uncharacterized.

Product Specs

Form
Lyophilized powder
Please note: We will prioritize shipping the format currently in stock. However, if you have specific requirements for the format, please indicate them in your order. We will prepare the product according to your request.
Lead Time
Delivery time may vary depending on the purchasing method or location. Please consult your local distributors for specific delivery times.
Note: All of our proteins are shipped with standard blue ice packs. If you require dry ice shipping, please inform us in advance as additional fees will apply.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.
Reconstitution
We recommend centrifuging the vial briefly before opening to ensure the contents settle at the bottom. Please reconstitute the protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL. We recommend adding 5-50% glycerol (final concentration) and aliquoting for long-term storage at -20°C/-80°C. Our standard final glycerol concentration is 50%. Customers can use this as a reference.
Shelf Life
The shelf life is influenced by various factors, including storage conditions, buffer composition, storage temperature, and the inherent stability of the protein.
Generally, the shelf life of the liquid form is 6 months at -20°C/-80°C. The shelf life of the lyophilized form is 12 months at -20°C/-80°C.
Storage Condition
Store at -20°C/-80°C upon receipt. Aliquoting is necessary for multiple uses. Avoid repeated freeze-thaw cycles.
Tag Info
Tag type is determined during the manufacturing process.
The specific tag type is determined during the production process. If you have a preferred tag type, please inform us, and we will prioritize its inclusion in the development process.

Target Background

Function
NDH-1 is an enzyme that transfers electrons from NADH, via FMN and iron-sulfur (Fe-S) centers, to quinones in the respiratory chain. In this species, the immediate electron acceptor for the enzyme is believed to be ubiquinone. NDH-1 couples the redox reaction with proton translocation (for every two electrons transferred, four hydrogen ions are translocated across the cytoplasmic membrane), effectively conserving the redox energy in a proton gradient.
