Description

Recombinant Production Strategies

HtpX homologs have been expressed in multiple systems, offering frameworks for B. bacilliformis applications:

Table 1: Expression Systems for HtpX Homologs

Host SystemSpeciesYield & PurityKey Parameters
E. coli BL21(DE3)B. henselae>90% purity (SDS-PAGE) Induction: 0.5 mM IPTG, 16–24 hr
Baculovirus/InsectB. bacilliformis (other proteins)High yields Temp: 21°C, Harvest: 120 hpi
Bacillus subtilisGut bacterium DX-3Active metalloprotease Induction: 1 mM IPTG, Cm resistance

Optimization Challenges

  • Solubility: Requires denaturing conditions (e.g., 6% Trehalose, Tris/PBS buffer) .

  • Tagging: His-tags facilitate purification via Ni-IDA affinity chromatography .

Pathogenicity Mechanisms

  • Host Interaction: HtpX homologs in B. henselae aid endothelial cell colonization and flea survival .

  • Complement Resistance: Autotransporter proteins (e.g., FhbpA) in B. bacilliformis bind factor H to evade immune detection .

Diagnostic and Therapeutic Potential

  • Antigenicity: Recombinant Bartonella proteins like Hbp/Pap31 show promise as vaccine candidates .

  • Enzymatic Activity: Neutral pH and heat resistance make HtpX suitable for industrial applications .

Research Gaps and Future Directions

  • Species-Specific Data: No direct studies on B. bacilliformis HtpX exist; current insights rely on homologs .

  • Structural Studies: Cryo-EM or X-ray crystallography is needed to resolve the catalytic mechanism .

  • In Vivo Models: Role in Carrion’s disease pathogenesis remains unexplored .

Product Specs

Form
Lyophilized powder
Note: We prioritize shipping the format currently in stock. If you have a specific format requirement, please indicate it when placing your order, and we will accommodate your request.
Lead Time
Delivery time may vary depending on the purchasing method or location. Please consult your local distributor for specific delivery timelines.
Note: All our proteins are shipped with standard blue ice packs. If you require dry ice shipping, please inform us in advance, as additional fees will apply.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.
Reconstitution
We recommend centrifuging the vial briefly before opening to ensure the contents settle at the bottom. Reconstitute the protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL. We suggest adding 5-50% glycerol (final concentration) and aliquoting for long-term storage at -20°C/-80°C. Our default glycerol concentration is 50%, which you can use as a reference.
Shelf Life
The shelf life is influenced by various factors, including storage conditions, buffer components, temperature, and the protein's inherent stability.
Generally, liquid form has a shelf life of 6 months at -20°C/-80°C. Lyophilized form has a shelf life of 12 months at -20°C/-80°C.
Storage Condition
Upon receipt, store at -20°C/-80°C. Aliquoting is necessary for multiple uses. Avoid repeated freeze-thaw cycles.
Tag Info
Tag type is determined during the manufacturing process.
The tag type will be determined during production. If you have a specific tag type requirement, please inform us, and we will prioritize developing the specified tag.
