Description

Introduction to Recombinant Bdellovibrio bacteriovorus Ribonuclease 3 (Rnc)

Recombinant Bdellovibrio bacteriovorus Ribonuclease 3 (Rnc) is an enzyme that belongs to the ribonuclease III family and is involved in the processing and maturation of RNA precursors into functional rRNA, mRNA, and other small RNAs . RNase III enzymes are double-strand-specific endoribonucleases that play a crucial role in gene expression regulation in bacteria .

Rnc in Bdellovibrio bacteriovorus

Bdellovibrio bacteriovorus is a predatory bacterium known for its ability to invade and consume other bacteria . Rnc in B. bacteriovorus contributes to its predatory lifestyle by:

  • Degradation of Host DNA During the predatory cycle, B. bacteriovorus secretes nucleases to degrade the host's DNA within the bdelloplast (the structure formed after invasion) .

  • Role in Host-Independent Growth Rnc is also expressed during the host-independent growth phase, suggesting its involvement in processing nucleic acids for nutrient acquisition or other metabolic processes .

Rnc and Antibiotic Resistance

Predatory bacteria like Bdellovibrio bacteriovorus offer a potential tool to combat antibiotic resistance . They can eliminate pathogens and reduce the pool of antibiotic resistance genes through the secretion of nucleases that degrade exogenous DNA .

Rnc in Salmonella enterica

  • Overexpression in Virulent Strains The rnc gene is overexpressed in high-virulence strains of Salmonella enterica .

  • Impact on Intracellular Survival Deletion of the rnc gene in Salmonella leads to reduced intracellular survival and replication rates in macrophage cells .

  • Modulation of Host Immune Response RNAs extracted from rnc knockout mutants induce higher levels of immune responses in the host, including innate immune factors like IFN-β, RIG-I, and MDA-5 . This is attributed to the accumulation of dsRNA in the absence of RNase III activity .

Methods for Studying Rnc

  • Immunoblotting Immunoblotting assays can be used to detect dsRNA accumulation in rnc knockout mutants .

  • Quantitative PCR (qRT-PCR) qRT-PCR is used to measure the expression levels of rnc and other genes throughout the bacterial life cycle .

  • DNase Assays DNase assays determine the extracellular DNase activity of bacterial strains with and without rnc .

  • RNase Activity Assays RNase activity assays assess the ability of RNC to degrade dsRNA .

Product Specs

Form
Lyophilized powder
Note: While we prioritize shipping the format currently in stock, please specify your format preference in order notes for customized fulfillment.
Lead Time
Delivery times vary depending on the purchasing method and location. Please contact your local distributor for precise delivery estimates.
Note: All proteins are shipped with standard blue ice packs. Dry ice shipping requires prior arrangement and incurs additional charges.
Notes
Avoid repeated freeze-thaw cycles. Store working aliquots at 4°C for up to one week.
Reconstitution
Centrifuge the vial briefly before opening to consolidate the contents. Reconstitute the protein in sterile, deionized water to a concentration of 0.1-1.0 mg/mL. For long-term storage, we recommend adding 5-50% glycerol (final concentration) and aliquoting at -20°C/-80°C. Our standard glycerol concentration is 50% and can serve as a reference.
Shelf Life
Shelf life depends on various factors including storage conditions, buffer composition, temperature, and protein stability. Generally, liquid formulations have a 6-month shelf life at -20°C/-80°C, while lyophilized forms have a 12-month shelf life at -20°C/-80°C.
Storage Condition
Store at -20°C/-80°C upon receipt. Aliquoting is essential for multiple uses. Avoid repeated freeze-thaw cycles.

Target Background

Function
This recombinant Bdellovibrio bacteriovorus Ribonuclease 3 (rnc) digests double-stranded RNA. It participates in processing the primary rRNA transcript to produce the immediate precursors of the large (23S) and small (16S) rRNAs. Additionally, it processes certain mRNAs and tRNAs encoded within the rRNA operon. If present in the organism, it also processes pre-crRNA and tracrRNA from type II CRISPR loci.
