GPR68 is implicated in tumor microenvironment (TME) acidosis, influencing cancer progression and metastasis. While most studies focus on human or murine GPR68, the bovine recombinant form serves as a comparative model for studying interspecies conservation of receptor function.
Key Research Findings
Proton Sensing and Signaling
GPR68 activates at pH <6.8, triggering Gαq/11-coupled pathways that elevate intracellular Ca²⁺ and activate protein kinase C (PKC) and mitogen-activated protein kinase (MAPK).
Structural studies using bovine GPR68 may elucidate conserved motifs critical for proton binding and G-protein coupling.
Tumor Suppressive vs. Oncogenic Roles
Tumor Suppression: Overexpression in prostate cancer cells inhibits migration without affecting proliferation.
Oncogenic Effects: Promotes angiogenesis and immune evasion in melanoma by inhibiting CD8⁺ T-cell and NK-cell infiltration.
Gender-Specific Modulation
GPR68 deficiency reduces melanoma growth in male mice but not females, correlating with enhanced immune cell infiltration (CD45⁺, CD8⁺ T cells, NK cells).
Bovine models could explore evolutionary conservation of these gender-dependent mechanisms.

Targeting GPR68 in Cancer Therapy
Radiosensitization
Inhibiting GPR68 (e.g., with OGM) enhances radiosensitivity by inducing ferroptosis via lipid peroxidation and iron dysregulation.
Bovine GPR68 models could validate species-specific responses to small-molecule inhibitors.
Immune Modulation
GPR68 antagonists may restore anti-tumor immunity by increasing CD8⁺ T-cell and NK-cell infiltration.
Bovine studies could inform cross-species immune checkpoint strategies.
Biomarker Potential
GPR68 expression correlates with molecular subtypes (e.g., triple-negative breast cancer).
Recombinant bovine GPR68 may aid in developing antibodies for diagnostic assays.
Challenges and Future Directions
Species-Specific Variability
Structural differences between bovine and human GPR68 may limit translational relevance.
Comparative studies using recombinant proteins could identify conserved motifs for drug design.
TME Complexity
Acidosis in tumors requires integration of GPR68 with other proton sensors (e.g., GPR4, GPR65).
Bovine models may clarify hierarchical roles of these receptors.
Gender Disparities
Mechanisms underlying male-specific GPR68 effects in melanoma warrant further investigation.
Product Specs
Form
Lyophilized powder Note: We will prioritize shipping the format currently in stock. However, if you have specific format requirements, please indicate them during order placement. We will fulfill your request to the best of our ability.
Lead Time
Delivery time may vary depending on the purchasing method or location. Please consult your local distributors for specific delivery timelines. Note: All our proteins are shipped with standard blue ice packs by default. If you require dry ice shipping, please contact us in advance, as additional fees will apply.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.
Reconstitution
We recommend centrifuging the vial briefly before opening to ensure the contents settle at the bottom. Reconstitute the protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL. We recommend adding 5-50% glycerol (final concentration) and aliquoting for long-term storage at -20°C/-80°C. Our default final glycerol concentration is 50%. Customers can use this as a reference.
Shelf Life
Shelf life is influenced by various factors, including storage conditions, buffer composition, storage temperature, and the inherent stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Storage Condition
Store at -20°C/-80°C upon receipt. Aliquoting is necessary for multiple uses. Avoid repeated freeze-thaw cycles.

GPR68 (G-protein coupled receptor 1) is a proton-sensing receptor involved in pH homeostasis. It may act as an osteoblastic pH sensor, regulating cell-mediated responses to acidosis in bone. GPR68 mediates its action by associating with G proteins, stimulating inositol phosphate (IP) production or Ca(2+) mobilization. The receptor exhibits minimal activity at pH 7.8 but is fully activated at pH 6.8. It also functions as a metastasis suppressor gene in prostate cancer.
