This kit employs a sandwich ELISA format, leveraging antibodies specific to the chicken FZD1 CRD .

Wnt Signaling Studies

  • Cardiac Hypertrophy: Recombinant FZD1 immunization in mice attenuated post-infarct hypertrophy by blocking Wnt/β-catenin signaling, reducing heart weight by 22% and improving ejection fraction .

  • Cancer: FZD1 overexpression correlates with drug resistance in acute myeloid leukemia (AML) and pancreatic ductal adenocarcinoma (PDAC). Silencing FZD1 reversed chemoresistance in NSCLC cells .

Developmental Biology

  • Embryogenesis: CRISPR-mediated FZD1 knockdown in chicken models disrupted axis formation and somite patterning, confirming its role in Wnt/planar cell polarity pathways .

Mechanistic Insights

  • Autoimmunization Strategy: Subcutaneous injection of recombinant FZD1 in mice generated anti-FZD1 antibodies that bound transmembrane FZD1, inhibiting Wnt signaling and hypertrophy .

  • Cross-Species Reactivity: Chicken FZD1 shares 97% sequence identity with bovine and 94% with human isoforms, enabling translational studies .

Challenges and Future Directions

  • Specificity: Off-target effects due to FZD1/FZD7 homology (74% sequence identity) require careful validation .

  • Therapeutic Potential: FZD1-targeted antibodies show promise in preclinical cardiac and cancer models but face hurdles in tissue specificity and immune tolerance .

Product Specs

Form
Lyophilized powder
Note: We prioritize shipping the format currently in stock. However, if you have specific format requirements, please indicate them when placing your order. We will accommodate your request if possible.
Lead Time
Delivery time may vary depending on the purchasing method and location. Please consult your local distributors for specific delivery timeframes.
Note: All our proteins are shipped with standard blue ice packs. If you require dry ice shipping, please inform us in advance. Additional fees will apply.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.
Reconstitution
We recommend centrifuging the vial briefly before opening to ensure the contents settle at the bottom. Reconstitute the protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL. For long-term storage, we recommend adding 5-50% glycerol (final concentration) and aliquoting at -20°C/-80°C. Our default glycerol concentration is 50%. Customers can use this as a reference.
Shelf Life
Shelf life is influenced by various factors, including storage conditions, buffer components, temperature, and the protein's inherent stability.
Generally, liquid form has a shelf life of 6 months at -20°C/-80°C. Lyophilized form has a shelf life of 12 months at -20°C/-80°C.
Storage Condition
Upon receipt, store at -20°C/-80°C. Aliquoting is necessary for multiple uses. Avoid repeated freeze-thaw cycles.

Target Background

Function
Frizzled-1 (FZD1) is a receptor for Wnt proteins. It plays a crucial role in the canonical Wnt/beta-catenin signaling pathway. This pathway leads to the activation of disheveled proteins, inhibition of GSK-3 kinase, nuclear accumulation of beta-catenin, and activation of Wnt target genes. A second signaling pathway involving PKC and calcium fluxes has been observed for certain family members. However, it remains unclear if this represents a distinct pathway or integrates with the canonical pathway. PKC appears to be required for Wnt-mediated inactivation of GSK-3 kinase. Both pathways seem to involve interactions with G-proteins. FZD1 may contribute to the transduction and intercellular transmission of polarity information during tissue morphogenesis or in differentiated tissues.
