Reconstitution
Centrifuge the vial briefly before opening to consolidate the contents. Reconstitute the protein in sterile deionized water to a concentration of 0.1-1.0 mg/mL. For long-term storage, we recommend adding 5-50% glycerol (final concentration) and aliquoting at -20°C/-80°C. Our standard glycerol concentration is 50%, which can be used as a reference.

ISU1 serves as a mitochondrial scaffold protein crucial for the de novo synthesis of iron-sulfur (Fe-S) clusters. These clusters are essential for the maturation of both mitochondrial and cytoplasmic [2Fe-2S] and [4Fe-4S] proteins. The process involves a two-step mechanism: (1) transient [2Fe-2S] cluster assembly on ISU1; (2) cluster transfer from ISU1 to a glutaredoxin, followed by mitochondrial [2Fe-2S] protein formation, [4Fe-4S] cluster synthesis, and targeted insertion into recipient apoproteins. This cluster assembly on ISU1 requires the cysteine desulfurase complex NFS1-ISD11 (sulfur donor), frataxin (putative iron donor), and an electron transfer chain (ferredoxin reductase and ferredoxin) that receives electrons from NADH.
