Description

Molecular Characterization of tspE

D. discoideum tspE is encoded by the gene DDB_G0291177 on chromosome 5 and belongs to a family of five tetraspanins (TspA-E) . Key features include:

FeatureDetail
Protein length231 amino acids (predicted molecular weight: ~26.5 kDa)
Structural motifsFour transmembrane domains, short extracellular loops (EC1/EC2), and a conserved "CCG" motif in EC2
Post-translational modificationsPotential N-glycosylation sites in EC1 and EC2
Gene expressionDetected in multicellular slug stages but not in vegetative amoebae

The "CCG" motif distinguishes tspE from other D. discoideum tetraspanins (e.g., TspA-D have "CCK/Y/C" motifs), aligning it more closely with metazoan tetraspanins .

Recombinant Production and Purification

Recombinant tspE is produced in heterologous systems for research applications:

Research Applications and Findings

Recombinant tspE facilitates studies on tetraspanin functions and interactions:

  • Localization: GFP-tagged Dictyostelium tetraspanins (including tspE homologs) localize to contractile vacuoles, suggesting roles in osmoregulation .

  • Functional insights: While tspE deletion strains show no overt developmental defects, its structural conservation with human tetraspanins (e.g., CD9/CD63) implies potential roles in membrane dynamics .

  • Biochemical tools: Recombinant tspE is used in ELISA and Western blotting to generate antibodies or study protein-protein interactions .

TspE’s unique CCG motif may enable interactions distinct from other family members, though functional studies remain limited .

Future Directions

Recombinant tspE provides a platform to explore:

  • Evolutionary conservation of tetraspanin functions across eukaryotes.

  • Structural basis of membrane protein assembly in contractile vacuoles.

  • Potential therapeutic targeting in diseases involving tetraspanins, such as cancer or parasitic infections .

Form
Lyophilized powder
Please note: We prioritize shipping the format currently in stock. However, if you have a specific format requirement, kindly indicate it in your order remarks, and we will prepare it accordingly.
Lead Time
Delivery time may vary depending on the purchasing method or location. For specific delivery timelines, please consult your local distributors.
Please note: Our proteins are shipped with standard blue ice packs by default. If you require dry ice shipping, please contact us in advance, as additional fees may apply.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.
Reconstitution
We recommend briefly centrifuging the vial prior to opening to ensure the contents are settled at the bottom. Reconstitute the protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL. For long-term storage, we recommend adding 5-50% glycerol (final concentration) and aliquoting at -20°C/-80°C. Our default final glycerol concentration is 50%. Customers can use this as a reference.
Shelf Life
Shelf life is influenced by various factors including storage conditions, buffer ingredients, storage temperature, and the inherent stability of the protein.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Storage Condition
Upon receipt, store at -20°C/-80°C. Aliquoting is necessary for multiple uses. Avoid repeated freeze-thaw cycles.
