Description

Signaling Mechanisms

The canine α1A-AR signals through:

  • Primary pathway: Gq/11 → Phospholipase C (PLC) → IP3/Ca²⁺ release

  • Secondary pathways:

    • Protein kinase C delta (PKCδ) activation → Troponin I phosphorylation

    • TRPC6 channel translocation → Enhanced calcium influx

Key functional differences from human α1A-AR include 12% divergence in extracellular loop sequences affecting ligand binding kinetics .

Cardiovascular Studies

  • Cardioprotection: Recombinant dog α1A-AR models show mitigated post-infarction mortality by suppressing RIP kinase-dependent necroptosis .

  • Inotropic Effects: Mediates positive inotropy in left ventricular cardiomyocytes through PKCδ-GLUT1/4 signaling .

Pharmacological Screening

Used to test subtype-specific agonists/antagonists:

CompoundEffect on Dog α1A-AREC50/IC50
A61603 (agonist)Full activation1.2 nM
DabuzalgronPartial agonism8.7 nM
Data extrapolated from murine/human studies

Clinical Implications

  • Heart Failure: α1A-AR activation reverses ventricular dysfunction in pressure-overload models by upregulating ERK1/2 survival pathways .

  • Drug Development: Subpressor doses of α1A agonists (e.g., A61603) prevent doxorubicin-induced cardiomyopathy in preclinical models .

Product Specs

Form
Lyophilized powder
Note: While we prioritize shipping the format currently in stock, we are happy to accommodate specific format requests. Please indicate your preference in the order notes, and we will do our best to fulfill your requirements.
Lead Time
Delivery time may vary depending on the purchase method and location. Please consult your local distributor for specific delivery estimates.
Note: All proteins are shipped with standard blue ice packs. If you require dry ice shipping, please inform us in advance as additional fees will apply.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.
Reconstitution
We recommend briefly centrifuging the vial before opening to ensure the contents settle at the bottom. Reconstitute the protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL. For long-term storage, we suggest adding 5-50% glycerol (final concentration) and aliquoting at -20°C/-80°C. Our standard glycerol concentration is 50%. Customers can use this as a reference.
Shelf Life
Shelf life depends on various factors, including storage conditions, buffer composition, temperature, and the protein's inherent stability.
Generally, liquid forms have a shelf life of 6 months at -20°C/-80°C. Lyophilized forms have a shelf life of 12 months at -20°C/-80°C.
Storage Condition
Upon receipt, store at -20°C/-80°C. Aliquot for multiple uses. Avoid repeated freeze-thaw cycles.
Tag Info
Tag type is determined during the manufacturing process.
The tag type is established during production. If you have a specific tag type in mind, please inform us, and we will prioritize its development.

Target Background

Function
This alpha-adrenergic receptor exerts its action by associating with G proteins, which activate a phosphatidylinositol-calcium second messenger system. Its effects are mediated through G(q) and G(11) proteins. Nuclear ADRA1A-ADRA1B heterooligomers regulate phenylephrine (PE)-stimulated ERK signaling in cardiac myocytes.
