Interaction with Cytoskeletal Proteins

ygfX binds to FtsZ and MreB, two essential cytoskeletal proteins in bacterial cell division. This interaction inhibits their polymerization:

  • FtsZ: GTP-dependent polymerization (critical for Z-ring formation during cell division) .

  • MreB: ATP-dependent polymerization (involved in cell shape maintenance) .

Role in Prodigiosin Biosynthesis

ygfX restores antibiotic production in Serratia strains with sdhE-ygfX deletions. Overexpression of ygfX, alongside CptB (its antitoxin), partially rescues prodigiosin synthesis, suggesting a regulatory link to metabolic pathways .

Membrane Localization and Multimer Formation

  • Localization: Primarily localized to the inner membrane .

  • Multimerization: Forms dimers or larger multimers via transmembrane helices. Critical residues include a periplasmic tryptophan and cytoplasmic aspartate .

Toxin-Antitoxin System Debate

  • Pro-Toxin View: Some studies classify ygfX as a membrane-bound toxin (CptA) that disrupts cell division, causing elongated or lemon-shaped cells .

  • Anti-Toxin Counterargument: Other research disputes this, emphasizing its interaction with SdhE (a FAD assembly factor) and its role in succinate dehydrogenase (SDH) activity regulation .

Recombinant Production in E. coli

ParameterDetailsSource
Expression SystemT7 promoter-driven (commonly in BL21(DE3) strains)
SolubilityExpressed in inclusion bodies; requires refolding or periplasmic secretion
ReconstitutionSuggested in sterile water (0.1–1.0 mg/mL) with 5–50% glycerol for stability

Key Challenges:

  • Toxicity: Overexpression may require specialized E. coli strains (e.g., C41/C43) to mitigate cell stress .

  • Storage Stability: Avoid repeated freeze-thaw cycles; store at -20°C/-80°C .

Potential Uses in Microbiology and Biotechnology

Outstanding Questions

  1. Dual Function: Does ygfX primarily act as a cytoskeletal inhibitor or SDH regulator?

  2. Conservation: Why is the sdhE-ygfX operon conserved across Enterobacteriaceae?

  3. Therapeutic Potential: Could ygfX derivatives inhibit bacterial pathogens by targeting cytoskeletal proteins?

Note: While we prioritize shipping the format currently in stock, please specify your format preference during order placement for customized preparation.
Lead Time
Delivery times vary depending on the purchasing method and location. Please contact your local distributor for precise delivery estimates.
Note: Our standard shipping includes blue ice packs. Dry ice shipping requires prior arrangement and incurs additional charges.
Notes
Avoid repeated freeze-thaw cycles. Store working aliquots at 4°C for up to one week.
Reconstitution
Centrifuge the vial briefly before opening to settle the contents. Reconstitute the protein in sterile deionized water to a concentration of 0.1-1.0 mg/mL. For long-term storage, we recommend adding 5-50% glycerol (final concentration) and aliquoting at -20°C/-80°C. Our default glycerol concentration is 50% and may serve as a reference.
Shelf Life
Shelf life depends on various factors including storage conditions, buffer composition, temperature, and protein stability. Generally, liquid formulations have a 6-month shelf life at -20°C/-80°C, while lyophilized formulations have a 12-month shelf life at -20°C/-80°C.
Storage Condition
Upon receipt, store at -20°C/-80°C. Aliquot for multiple uses. Avoid repeated freeze-thaw cycles.

Target Background

Function

Recombinant Escherichia coli Uncharacterized protein ygfX (ygfX)

YgfX is a putative inner membrane protein. Studies have indicated that it is non-toxic, exhibiting no effects on growth in LB or minimal media up to 6 and 21 hours post-induction, respectively. It interacts with the cytoskeletal proteins FtsZ and MreB, inhibiting FtsZ GTP-dependent polymerization and MreB ATP-dependent polymerization. Furthermore, ygfX restores prodigiosin antibiotic (Pig) production in Serratia strains with sdhE-ygfX deletions. Overexpression of ygfX and CptB also partially restores Pig production in Serratia.
