Reconstitution
Centrifuge the vial briefly before opening to collect the contents. Reconstitute the protein in sterile deionized water to a concentration of 0.1-1.0 mg/mL. For long-term storage, we recommend adding 5-50% glycerol (final concentration) and aliquoting at -20°C/-80°C. Our default glycerol concentration is 50% and can serve as a reference.

Shelf life depends on various factors including storage conditions, buffer composition, temperature, and protein stability. Generally, liquid formulations have a 6-month shelf life at -20°C/-80°C, while lyophilized forms have a 12-month shelf life at -20°C/-80°C.

Target Background

Function
NLRC3 is a negative regulator of the innate immune response. It attenuates signaling pathways activated by Toll-like receptors (TLRs) and the DNA sensor STING/TMEM173 in response to pathogen-associated molecular patterns (PAMPs), such as intracellular poly(dA:dT) (but not poly(I:C)), or DNA virus infection (e.g., Herpes simplex virus 1, HSV1). It may modulate TLR4 signaling by affecting TRAF6 ubiquitination, reducing activating Lys-63-linked ubiquitination while leaving degradative Lys-48-linked ubiquitination unaffected. NLRC3 inhibits the PI3K-AKT-mTOR pathway, potentially by directly interacting with the PI3K regulatory subunit p85 (PIK3R1/PIK3R2), disrupting its association with the catalytic subunit p110 (PIK3CA/PIK3CB/PIK3CD), and reducing p85 activation. Through its regulation of the PI3K-AKT-mTOR pathway, NLRC3 controls cell proliferation, predominantly in intestinal epithelial cells. It may also influence NOD1- or NOD2-mediated NF-κB activation and inflammatory responses by preventing NLRP3 inflammasome formation, CASP1 cleavage, and IL-1β maturation.
