Characteristics of Acylphosphatase

Lactobacillus plantarum is a lactic acid bacterium with the ability to express enzymes with therapeutic potential when produced recombinantly . Acylphosphatases (AcyPs) are small, highly conserved enzymes that catalyze the hydrolysis of acylphosphates .

Applications in Immunological Research

Recombinant L. plantarum can be engineered to express and deliver antigens, enhancing immune responses. For instance, recombinant L. plantarum expressing the H7N9 antigen has been shown to induce IgA expression in the lungs and intestines of mice, with the HA1-DCpep group demonstrating a more pronounced advantage .

Metabolic Effects

Research indicates that Lactobacillus plantarum influences metabolic pathways. For example, in Streptococcus bovis, inactivation of ccpA affects glycolysis and pyruvate metabolism, with genes encoding acetyl-CoA carboxylase biotin and acyl phosphatase being downregulated in the ccpA mutant .

Treatment of Hypertension

Recombinant Lactobacillus plantarum expressing angiotensin-converting enzyme inhibitory peptides (ACEIPs) has demonstrated effectiveness in treating hypertension in spontaneously hypertensive rats (SHRs) . Oral administration of recombinant Lb. plantarum NC8 significantly decreased systolic blood pressure, increased nitric oxide levels, and decreased endothelin and angiotensin II levels . Reconstitution
Centrifuge the vial briefly before opening to settle the contents. Reconstitute the protein in sterile deionized water to a concentration of 0.1-1.0 mg/mL. We recommend adding 5-50% glycerol (final concentration) and aliquoting for long-term storage at -20°C/-80°C. Our standard glycerol concentration is 50% and can serve as a reference.
Shelf Life
Shelf life depends on several factors including storage conditions, buffer composition, temperature, and the protein's inherent stability. Generally, liquid formulations have a 6-month shelf life at -20°C/-80°C, while lyophilized formulations have a 12-month shelf life at -20°C/-80°C.
Storage Condition
Upon receipt, store at -20°C/-80°C. Aliquoting is essential for multiple uses. Avoid repeated freeze-thaw cycles.
