Description

Production and Purification

Recombinant Mouse VIP36-like protein is synthesized using multiple expression systems:

Host SystemPurityTagApplications
E. coli≥85%None/His-tagSDS-PAGE, WB, ELISA
Yeast/Baculovirus≥85%VariableStructural studies
Mammalian Cells>95%His-tagFunctional assays, immunoprecipitation

Glycoprotein Trafficking:

  • Regulates ER export of select glycoproteins by interacting with ERGIC-53, a cargo receptor .

  • Loss of LMAN2L disrupts surface expression of integrin α6 (ITGA6), suggesting a role in vesicular transport .

Viral Interactions:

  • Human cytomegalovirus (HCMV) protein US2 targets LMAN2L for TRC8-mediated degradation to impair host immune responses .

  • Associates with enterovirus 3A protein, SCAMP3, and PI4KIIIβ to form replication complexes, enhancing viral RNA synthesis .

Sugar-Binding Activity:

  • Demonstrates calcium-dependent lectin activity for high-mannose glycans, critical for quality control in the ER .

Study FocusKey FindingsReference
Lectin ActivityConfirmed sugar-binding specificity for Manα1-2Manα1-2Man motifs using SPR and flow cytometry
Viral ReplicationIdentified SCAMP3-LMAN2L-PI4KIIIβ complex as critical for enterovirus replication
Host-Pathogen InteractionHCMV US2 degrades LMAN2L to reduce ITGA6 surface expression, evading immunity
Structural AnalysisRecombinant protein used to map ER retention signals and glycosylation sites

Research Applications

  • Glycobiology: Used to analyze mannose-binding specificity and ER-Golgi transport mechanisms .

  • Virology: Employed in studies on enterovirus and HCMV replication strategies .

  • Protein Interaction Mapping: Facilitates identification of binding partners (e.g., SCAMP3, PI4KIIIβ) via co-immunoprecipitation .

Challenges and Future Directions

  • Functional Redundancy: VIP36-like proteins share overlapping roles with VIP36, complicating phenotype analysis .

  • Therapeutic Potential: Targeting LMAN2L-pathogen interactions (e.g., with HCMV or enteroviruses) could yield antiviral strategies .

Form
Lyophilized powder
Note: We will prioritize shipping the format currently in stock. However, if you have a specific format requirement, please indicate it during order placement, and we will fulfill your request.
Lead Time
Delivery time may vary depending on the purchasing method or location. Please consult your local distributors for specific delivery details.
Note: All of our proteins are shipped with standard blue ice packs. If dry ice shipping is required, please contact us in advance, as additional fees will apply.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.
Reconstitution
We recommend briefly centrifuging the vial before opening to ensure the contents settle at the bottom. Reconstitute the protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL. We recommend adding 5-50% glycerol (final concentration) and aliquoting for long-term storage at -20°C/-80°C. Our default glycerol final concentration is 50%, which can be used as a reference.
Shelf Life
Shelf life is influenced by various factors, including storage conditions, buffer composition, temperature, and the intrinsic stability of the protein.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Storage Condition
Upon receipt, store at -20°C/-80°C. Aliquoting is recommended for multiple uses. Avoid repeated freeze-thaw cycles.
