Description

Functional Role

  • Energy Transfer:

    • Facilitates energy transfer from phycocyanin (PC) to AP core components, ultimately directing light energy to photosystem II (PSII) .

    • Interacts with linker proteins (e.g., apcE, cpcG) to stabilize phycobilisome architecture .

  • Biosynthesis:

    • Requires bilin lyases (CpcS-I/CpcU) for PCB attachment to cysteine residues Cys-81 and Cys-82 .

    • ApcD is synthesized as an apoprotein and undergoes chromophorylation post-translationally .

Heterologous Expression and Applications

  • Production in E. coli:

    • Recombinant apcD is expressed alongside ApcB to form functional AP-B complexes, yielding ~3–12 mg/L holoprotein in optimized systems .

    • Requires co-expression of CpcS-I/CpcU for proper PCB attachment .

  • Applications:

    • Used as a fluorescent probe in biomedical imaging due to its high quantum yield .

    • Exhibits antioxidant activity, correlating with chromophore binding efficiency .

Interactions and Functional Partners

  • Core Phycobilisome Components:

    • Binds to ApcB (β subunit) to form AP-B .

    • Interacts with apcE (core linker protein) and cpcG (rod-core linker) for energy transfer optimization .

  • Regulatory Proteins:

    • Associates with LcpA, a novel linker mediating phycobilisome-PSII interactions .

Research Challenges and Advances

  • Chromophore Stability:

    • ApcD’s PCB-binding efficiency varies with expression conditions, influencing fluorescence properties .

  • Structural Insights:

    • Cryo-EM studies reveal apcD’s role in phycobilisome-PSII docking, though its exact binding motifs remain under investigation .

Product Specs

Form
Lyophilized powder. We will preferentially ship the format we have in stock. If you have special format requirements, please note them when ordering, and we will fulfill your request.
Lead Time
Delivery time varies depending on the purchase method and location. Consult your local distributors for specific delivery times. All proteins are shipped with standard blue ice packs. For dry ice shipping, contact us in advance; additional fees apply.
Notes
Avoid repeated freezing and thawing. Store working aliquots at 4°C for up to one week.
Reconstitution
Briefly centrifuge the vial before opening to collect contents at the bottom. Reconstitute the protein in sterile deionized water to a concentration of 0.1-1.0 mg/mL. We recommend adding 5-50% glycerol (final concentration) and aliquoting for long-term storage at -20°C/-80°C. Our default final glycerol concentration is 50% for your reference.
Shelf Life
Shelf life depends on several factors: storage conditions, buffer components, storage temperature, and protein stability. Generally, the liquid form has a shelf life of 6 months at -20°C/-80°C, while the lyophilized form has a shelf life of 12 months at -20°C/-80°C.
Storage Condition
Store at -20°C/-80°C upon receiving. Aliquot for multiple uses. Avoid repeated freeze-thaw cycles.
