Notes
Avoid repeated freezing and thawing. Store working aliquots at 4°C for up to one week.
Reconstitution
Briefly centrifuge the vial before opening to collect the contents at the bottom. Reconstitute the protein in sterile deionized water to a concentration of 0.1-1.0 mg/mL. We recommend adding 5-50% glycerol (final concentration) and aliquoting for long-term storage at -20°C/-80°C. Our default final glycerol concentration is 50% for your reference.
Shelf Life
Shelf life depends on several factors, including storage conditions, buffer components, storage temperature, and protein stability. Generally, the shelf life for the liquid form is 6 months at -20°C/-80°C, and for the lyophilized form, it is 12 months at -20°C/-80°C.

Target Background

Function
Plays a central role in the 2-thiolation of mcm(5)S(2)U at tRNA wobble positions of tRNA(Lys), tRNA(Glu), and tRNA(Gln). It likely functions by forming a heterodimer with ncs6, which transfers sulfur from thiocarboxylated urm1 onto the uridine of tRNAs at the wobble position. Prior mcm(5) tRNA modification by the elongator complex is necessary for 2-thiolation. May also be involved in protein urmylation.
