Description

pH Regulation in the Central Nervous System

Recombinant Rat CA4 contributes to extracellular buffering in the hippocampus by modulating alkaline transients during neuronal activity. Knockout (KO) mouse models revealed:

  • CA4 and CA XIV redundantly regulate interstitial pH, with CA4 accounting for ~60% of buffering capacity .

  • Double KO mice (CA4/CA14) exhibit amplified alkaline shifts (2-fold increase vs. WT), confirming synergistic roles .

Renal and Pulmonary Functions

  • Anchored to luminal surfaces of renal tubules and pulmonary capillaries via GPI linkage, CA4 facilitates bicarbonate transport and CO₂ excretion .

  • In renal proximal tubules, it enhances AE3-mediated Cl⁻/HCO₃⁻ exchange, critical for maintaining acid-base balance .

Disease Modeling

  • Retinitis Pigmentosa (RP17): Mutations in CA4 disrupt retinal pH homeostasis, leading to photoreceptor degeneration .

  • Metabolic Acidosis: Impaired CA4 activity correlates with bicarbonate transport defects in renal tubules .

Challenges and Future Directions

Current limitations include the lack of full-length protein constructs in commercial preparations . Future studies could explore:

  • Structural modifications to enhance stability for in vivo delivery.

  • CRISPR-edited CA4 models to dissect its role in neurodegenerative diseases .

Product Specs

Form
Lyophilized powder. We will ship the format we have in stock. If you have special format requirements, please note them when ordering, and we will accommodate your request.
Lead Time
Delivery times vary based on purchasing method and location. Contact your local distributor for specific delivery times. All proteins are shipped with standard blue ice packs. For dry ice shipping, please contact us in advance; extra fees apply.
Notes
Avoid repeated freezing and thawing. Working aliquots can be stored at 4°C for up to one week.
Reconstitution
Briefly centrifuge the vial before opening to collect contents at the bottom. Reconstitute the protein in sterile deionized water to a concentration of 0.1-1.0 mg/mL. We recommend adding 5-50% glycerol (final concentration) and aliquoting for long-term storage at -20°C/-80°C. Our default final glycerol concentration is 50%.
Shelf Life
Shelf life depends on storage conditions, buffer components, storage temperature, and protein stability. Liquid form shelf life is generally 6 months at -20°C/-80°C. Lyophilized form shelf life is generally 12 months at -20°C/-80°C.
Storage Condition
Store at -20°C/-80°C upon receiving. Aliquot for multiple uses. Avoid repeated freeze-thaw cycles.
Tag Info
The tag type will be determined during the manufacturing process. If you have a specific tag type requirement, please inform us, and we will prioritize developing the specified tag.

Target Background

Function
Reversibly hydrates carbon dioxide. May stimulate the sodium/bicarbonate transporter activity of SLC4A4.
Gene References Into Functions
1. Blocking endothelin receptors can cause vascular mineral loss, which appears dependent on membrane-bound carbonic anhydrase IV activation. This mineral loss may occur via local pH changes similar to those observed in bone resorption. (PMID: 16144999)

Subcellular Location
Cell membrane; Lipid-anchor, GPI-anchor.
Tissue Specificity
Present in kidney and lung. Also particularly abundant in brain, muscle, heart and liver. Not detected in skin or spleen.
