Description

Production and Purification

Recombinant TMBIM4 is synthesized using E. coli expression systems, ensuring high purity and consistency . Key production details include:

ParameterDetail
Expression SystemE. coli (in vitro)
Purity≥95% (SDS-PAGE verified)
StorageLyophilized or liquid form

This method avoids eukaryotic post-translational modifications, simplifying structural studies while preserving core functional domains .

Anti-Apoptotic Activity

TMBIM4 inhibits apoptosis induced by intrinsic (e.g., Bax overexpression) and extrinsic (e.g., TNFα/Fas ligand) stimuli. It modulates:

  • Calcium signaling: Regulates inositol 1,4,5-trisphosphate (IP3)-mediated Ca²⁺ release and capacitative Ca²⁺ entry .

  • ER stress response: Retains pro-survival activity under cellular stress .

Interaction Network

Key molecular partners identified via affinity chromatography and STRING-db analysis :

Interacting ProteinRoleInteraction Score
PMLScaffolds PML nuclear bodies0.898
SIGMAR1Lipid transport regulation0.876
ITPR3Mediates IP3 calcium release0.814

Experimental Use Cases

  • Apoptosis assays: Used to study Bax-driven cell death in cancer models .

  • Calcium flux studies: Employed in IP3 receptor modulation experiments .

  • Structural biology: Serves as a template for transmembrane protein crystallization trials .

Limitations

  • Lack of native glycosylation in prokaryotic systems may affect ligand binding studies .

  • Functional redundancy with paralogs (TMTC1-3) complicates phenotype interpretation .

Evolutionary and Clinical Relevance

  • Conservation: Orthologs exist in mammals, reptiles, amphibians, and invertebrates but absent in plants/fungi .

  • Disease associations: Preliminary links to cancer progression and chemoresistance, though less characterized than TMBIM6 .

Product Specs

Form
Lyophilized powder
Note: While we prioritize shipping the format currently in stock, we can accommodate specific format requests. Please indicate your preference in the order notes, and we will prepare accordingly.
Lead Time
Delivery times may vary depending on the purchase method and location. Please consult your local distributor for specific delivery information.
Note: All proteins are shipped with standard blue ice packs. If you require dry ice shipping, please contact us in advance for an additional fee.
Notes
Repeated freezing and thawing is discouraged. For optimal results, store working aliquots at 4°C for up to one week.
Reconstitution
Prior to opening, we recommend briefly centrifuging the vial to ensure the contents settle at the bottom. Reconstitute the protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL. We recommend adding 5-50% glycerol (final concentration) and aliquoting for long-term storage at -20°C/-80°C. Our standard final glycerol concentration is 50%. Customers can use this as a reference.
Shelf Life
Shelf life is influenced by factors including storage conditions, buffer components, temperature, and the protein's inherent stability.
Generally, liquid form has a 6-month shelf life at -20°C/-80°C. Lyophilized form maintains a 12-month shelf life at -20°C/-80°C.
Storage Condition
Upon receipt, store at -20°C/-80°C. Aliquoting is recommended for multiple uses. Avoid repeated freeze-thaw cycles.
Tag Info
Tag type is determined during the manufacturing process.
The tag type is established during production. If you have a specific tag type preference, please inform us, and we will prioritize its development.
