Function
Endoribonuclease piwil2 is crucial for spermatogenesis by repressing transposable elements, ensuring germline integrity. It's essential for meiotic differentiation of spermatocytes, germ cell differentiation, and spermatogonial stem cell self-renewal. Piwil2 participates in the piRNA metabolic process, which represses transposable elements during meiosis via piRNA-Piwi protein complexes, leading to transposon methylation and repression. In piRNA biosynthesis, piwil2 is key in the ping-pong amplification cycle. As a 'slicer-competent' endoribonuclease, it cleaves primary piRNAs, which load onto 'slicer-incompetent' piwil4. Piwil2 slicing creates a pre-miRNA intermediate (processed into mature piRNAs) and a degraded 16-nucleotide by-product. It's required for piwil4/miwi2 nuclear localization and association with antisense secondary piRNAs. Piwil2 also represses circadian rhythms by promoting ARNTL/BMAL1 and CLOCK stability and activity.
