Function
This antibody targets a tri-functional enzyme essential for RNA repair and tRNA splicing. Specifically, it repairs RNA breaks with 2',3'-cyclic phosphate and 5'-OH ends through three sequential enzymatic activities: (1) opening of the 2',3'-cyclic phosphodiester bond (CPDase activity), (2) phosphorylation of the 5' terminus (kinase activity), and (3) ligation of the RNA fragments (ligase activity). While proficient at sealing pre-adenylylated AppRNA(OH), its activity is deficient in transferring AMP to pRNA(OH) to form AppRNA(OH). The CPDase and kinase reactions are relatively insensitive to RNA length; however, ligase activity decreases with shorter RNA. This enzyme also exhibits DNA splicing capabilities, specifically for DNA with a single 3'-terminal ribonucleoside 2',3'-cyclic-PO(4). It interacts with various RNA species, including mRNA and both mature and immature tRNA. Its role in tRNA splicing is crucial for precursor tRNA maturation. In vitro studies demonstrate its ability to circularize introns cleaved from pre-tRNA. The enzyme's function appears unrelated to the unfolded protein response (UPR) in the endoplasmic reticulum. Furthermore, it plays a significant role in auxin signaling and polar auxin transport during organ morphogenesis.

Mainly expressed in proliferating cells and tissues such as root meristems, the vasculature of developing plantlets, flowers and elongating tissue.
